ABSTRACT

The objective of this study was to investigate the effects of a gel formulation from the association of Plectranthus neochilus and Cnidoscolus quercifolius on tissue repair in cutaneous wounds in rats. A surgical wound was induced in 35 Wistar rats and treated according to group: G1 - commercial phytotherapeutic gel; G2 - Carbopol gel 1%; G3, 4, 5 - gel formulation from Boldo-gambá and Favela (FGBF) at 2.5%, 5%, and 10%, respectively. 1ml of the product was applied topically daily, for 14 days. Macroscopic evaluation of the wound showed inflammation, granulation, and epithelization in all groups. The FGBF 2.5% group showed greater angiogenic potential. There was a significant difference between the surgical area of the wounds treated with FGBF 2.5%, 5%, or 10% compared to the group with the commercial phytotherapeutic gel. On histomorphometry of the skin, there were reepithelization of the epidermis and superficial dermis, longitudinal collagen fibers, fibroblasts, and blood vessels, and in the deeper dermis, fibroblasts, transverse and longitudinal collagen fibers, blood vessels, and inflammatory cells. The 2.5% formulation had the greatest increase in fibroblast proliferation and most intense collagenization on day 14 of treatment.(AU)


Este trabalho objetivou investigar os efeitos de uma formulação em gel da associação da Plectranthus neochilus e da Cnidoscolus quercifolius no processo de reparação tecidual em feridas cutâneas de ratos. Foi induzida uma ferida cirúrgica em 35 ratos Wistar, sendo tratadas de acordo com os grupos: G1 - fitoterápico comercial; G2 - gel de carbopol 1%; G3, G4 e G5 - formulação gel boldo-gambá mais favela (FGBF) 2,5%, 5% e 10%, respectivamente. Aplicou-se 1mL do produto, via tópica, diariamente, durante 14 dias. Na avaliação macroscópica das feridas, verificou-se inflamação, granulação e epitelização em todos os grupos. O grupo FGBF 2,5% apresentou maior potencial angiogênico. Houve diferença significativa entre as áreas cirúrgicas das feridas tratadas com os FGBF 2,5%, 5% ou 10%, comparados ao grupo com o gel fitoterápico comercial. Na histomorfometria da pele, observou-se reepitelização da epiderme e da derme superficial, fibras colágenas longitudinais, fibroblastos e vasos sanguíneos e, na derme profunda, fibroblastos, fibras colágenas transversais e longitudinais, vasos sanguíneos e células inflamatórias. A formulação a 2,5% teve o maior aumento na proliferação de fibroblastos e mais intensa colagenização no dia 14 de tratamento.(AU)

ABSTRACT

Avaliaram-se dois protocolos para a produção de plasma rico em plaquetas (PRP) com o sangue de 20 cães adultos. Foram coletados três frascos de sangue em que um deles foi usado para produção do PRP por meio do protocolo A - centrifugação única a 1200rpm/10min -, o outro para fabricação do PRP pelo protocolo B - primeira centrifugação a 1200rpm/10min e a segunda centrifugação a 1600rpm/10min - e o terceiro para realização da contagem plaquetária no sangue total, que serviu de parâmetro para os valores alcançados no PRP. O protocolo no qual foi possível alcançar maior concentração plaquetária foi testado em outros 20 cães para avaliar sua reprodutibilidade. Constatou-se que o protocolo B resultou em maior plaquetometria em 100 por cento das amostras e concluiu-se ser ele eficiente para a produção do PRP em cães.


The objective of this paper was to analyze two protocols for the production of platelet rich plasma (PRP) in dogs. Peripheral blood of 20 adult dogs was collected into three tubes. The first was processed through protocol A - single centrifugation at 1200rpm for 10min - , the second was submitted to protocol B - a first centrifugation at 1200rpm for 10min and a second centrifugation at 1600rpm for 10min - and the third was used to perform platelet count in whole blood, which served as a parameter for values obtained in PRP. The protocol in which it was possible to achieve a higher platelet count was tested in other 20 dogs to evaluate its reproducibility. Protocol B resulted in a superior platelet count in 100 percent of the samples, concluding that the referred protocol is effective for PRP production in dogs.

ABSTRACT

The serum levels of Gamma-GT in dogs naturally infected by Leishmania (Leishmania) chagasi were evaluated. Two groups of animals were used: the first with 41 naturally infect dogs and the second with 14 uninfect animals. The results showed 17.1% (7/41) dogs presented high levels of gamma-GT, but no difference between groups was observed. So, the serum level of gamma-GT is not a tool for diagnosis of visceral leishmaniasis in dogs.

ABSTRACT

Aiming to evaluating some conditions of EDTA 10% serum and plasma storage for urea and creatinine values determinations in canines as well as analyzing the possibility of serum substitution by plasma, a total of 3600 samples (1800 serum and 1800 of plasma) were analyzed at room, refrigeration and freezing temperatures. Different time intervals were used in the analysis: immediately after sampling (time 0), and at 2, 6, 12, 24, and 72 hours, and 30 and 60 days after the blood collection. The data showed that urea remains conserved for more time in the refrigerated serum sample until 30 days and the creatinine in the frozen serum sample until 72 hours. Serum showed more stability than plasma in all storage conditions established.


O trabalho teve como objetivos avaliar algumas condições de estocagem de soro e de plasma com EDTA 10% utilizados para determinação dos valores de uréia e creatinina em cães, e avaliar a possibilidade de substituir o soro pelo plasma conservado com EDTA 10%. Foram realizadas 3600 análises (1800 amostras de soro e 1800 de plasma) com amostras que permaneceram em temperaturas ambiente, de refrigeração e de congelamento. As análises foram realizadas imediatamente após a obtenção da amostra (tempo 0) e após 2, 6, 12, 24, 72 horas e 30 e 60 dias após a coleta do sangue. Os resultados permitiram concluir que a uréia conservou-se por mais tempo na amostra sérica refrigerada até 30 dias e a creatinina na amostra sérica congelada até 72h. O soro mostrou mais estabilidade que o plasma em todas as condições de estoque estabelecidas.

ABSTRACT

Thirteen clinicaly healthy crossbred (Holstein x Zebu) aged between 20 and 45 months, raised in Rio de Janeiro State, Brazil, were utilized. Samples of blood (2), without anticoagulant, were colected from each animal by jugular vein puncture. It was determined the values for total protein, albumin (commercial kits), globulins and albumin/globulins ratio by mathematics calculation. The observed limits, mean and respective standard deviation in g/dl were: 5.70 to 8.80 and 7.01 more or less 0.65 (total protein); 2.60 to 4.20 and 3.10 more or less 0.36 (albumin); 2.30 to 5.00 and 3.70 more or less 0.59 (globulins) and 0.60 to 3.50 and 2.25 more or less 0.87 (albumin/globulins ratio)
